چکیده

Although it is well established that only two enzymes, transamidinase and guanidoacetate methylpherase, are directly involved in the formation of creatine (l-4), almost nothing is known about the regulation of creatine synthesis. Since various diseases alter creatine metabolism (5), it is possible that a study of transamidinase and guanidoacetate methylpherase activities in some of these diseases would furnish clues to the physiological control of creatine synthesis. One of the diseases that disturbs creatine metabolism is hyperthyroidism (6) and it will be shown in the present report that hyperthyroidism reduces kidney transamidinase activity. Since this could be due to the elevated blood creatine concentration of hyperthyroidism (B), creatine feeding experiments were conducted. The data obtained show that excess creatine also reduces the activity of kidney transamidinase in the rat and demonstrate a feedback mechanism (7) in a mammal.

The mechanism of kidney transamidinase reduction in vitamin E-deficient rabbits.

A recent investigation by Van Pilsum and Wahman (1) revealed low kidney transamidinase activities in vitamin Edeficient rabbits. One logical mechanism to explain this finding is suggested by the observation that dietary creatine depresses kidney transamidinase activity in rats (2, 3). I f the creatinuria of vitamin E deficiency depresses glycocyamine synthesis by kidney transamidinase while cre...

Transamidinase Activities in Vitro of Kidneys from Tumor-bearing Mice and Rats Fed Diets Supplemented with Protein or Certain Amino Acids.

Some aspects of the alteration of kidney transamidinase activities by rapidly growing tumors have been investigated. Transamidinase activities, in vitro, were determined in kidneys from normal and tumor-bearing animals fed diets supple mented individually with arginine, glycine, creatine, and protein. These dietary constituents were chosen because each had been previously reported to have a gre...

The effect of growth hormone and thyroxine on the amount of L-arginine:glycine amidinotransferase in kidneys of hypophysectomized rats. Purification and some properties of rat kidney transamidinase.

The initial step in creatine biosynthesis involves the transfer of an amidine group from arginine to glycine to form guanidinoacetic acid and is catalyzed by the enzyme L-arginine:glycine amidinotransferase (transamidinase). A procedure for the purification of rat kidney transamidinase by chromatography with DEAEcellulose and phenyl-Sepharose is described in this report. Two fractions of enzyme...

Transamidination in the nephrectomized rat.

Reaction 1 is catalyzed by the enzyme arginine-glycine transamidinase (l), and Reaction 2 by guanidinoacetic acid methylpherase (2). Until recently, the only site of the enzyme transamidinase in mammals was thought to be the mammalian kidney (1, 3). In brief footnotes, Horner et al. (4) mentioned preliminary experiments which indicated that an enzyme which catalyzes the reversal of Reaction 1 i...

Transamidinase activities, in vitro, of kidneys from rats fed diets supplemented with nitrogen-containing compounds.

It has been reported by Walker (1) and Fitch et al. (2) that kidneys from rats fed normal diets supplemented with creatine had lower transamidinase activities in vitro than kidneys from rats fed normal diets. It therefore seemed of interest to determine if the creatine-depressing effect could be prevented or reversed by the addition of certain compounds to creatine-containing diets. Also, it wa...
